Making X-gal Plates

Stock X-gal = 10% in DMF

Stock 1.5% “Top Agar” 100 ml in 250 ml bottle

Method 1 (spread onto LB plates)

For each large plate:

25 ul X-gal stock + 100 ul LB (use glass if possible)

Spread onto large plate and let dry

Method 2 (“Top Agar”)

Microwave 100 ml of 1.5% agar in LB

Cool to ~ 57 deg and add:

200 ul 10% X-gal stock

ampicilin to 100 ug/ml

Add 10 ml per large plate (5 ml per small plate)

You can add to top of existing plate or make a fresh plate

Let solidify
